FDA Grants Priority Review to Pharming’s Joenja for Children With APDS

Pharming (Euronext: PHARM / Nasdaq: PHAR) announced today, September 25, 2026, that the U.S. Food and Drug Administration has accepted its supplemental New Drug Application (sNDA) for lower doses of Joenja (leniolisib) and granted the filing Priority Review, with a PDUFA target action date of January 30, 2027. The application seeks approval for children aged 4 and older who weigh 13 kg to 27 kg and have APDS (activated phosphoinositide 3-kinase delta syndrome), a rare primary immunodeficiency.

This extends Joenja’s reach to smaller children. The oral selective PI3K-delta inhibitor was recently approved for children aged 4 to 11 with APDS who weigh at least 27 kg, and the new sNDA would open the door to the 13–27 kg group. The filing is backed by positive data from an open-label, multinational, single-arm Phase III study in children aged 4 to 11: over 12 weeks, researchers saw improvement in two hallmarks of the disease — reduced lymphadenopathy and increased naive B cells — indicating correction of the underlying immune defect, with additional clinical pharmacology assessments supporting dosing in the lower-weight group.

Why it matters: pediatric label expansions are one of the most reliable ways a rare-disease company can widen the addressable market for an already-approved drug, and every additional weight cohort compounds Joenja’s franchise value. Priority Review — granted when a medicine, if approved, would offer a significant improvement in the effectiveness or safety of treatment for a serious condition — also compresses the waiting game to roughly six months instead of the standard ten, giving investors a concrete January 2027 catalyst to trade around.

What to watch: the January 30, 2027 PDUFA date is the obvious milestone, but pay attention to the final label language and how quickly payers cover the lower-dose formulation once (and if) it is approved.
