What happened
Kodiak Sciences (KOD) soared roughly 65–70% to about $54.50 in premarket trading Monday after reporting that both of its experimental eye drugs met the primary goal of the DAYBREAK Phase 3 trial — putting the stock on track for its best single-day gain on record and its highest level since February 2022.
The trial tested Zenkuda and KSI-501 in wet age-related macular degeneration, a leading cause of vision loss in older adults. Instead of two costly separate studies, Kodiak combined both drugs into one trial with a shared control group, testing against Regeneron’s Eylea, the current standard of care. Both drugs proved non-inferior to Eylea at restoring vision after one year of treatment, and Zenkuda showed what the company called “a potential new standard-of-care profile,” with relief immediately after administration and durable benefit.
- Move: +65–70% premarket to ~$54.42–$54.84 (sources: Barron’s, CoinCentral); Friday close $32.35
- DAYBREAK: both drugs non-inferior vs. Regeneron Eylea on one-year vision gain
- Next catalyst: Biologics License Application for Zenkuda planned in Q4 2026; PEAK MESI data Q4 2026, PINNACLE MESI data Q2 2027
Why it matters
A double win on a single trial is rare in biotech and derisks Kodiak’s whole pipeline — a first product to market would validate the company’s antibody platform and open a share of the estimated $15 billion global retinal vascular disease drug market. Commercial success now hinges on differentiation: extended durability and longer dosing intervals versus Eylea, not just non-inferiority. The earlier GLOW2 win in diabetic retinopathy (62.5% vs. 3.3% sham on DRSS improvement) already had UBS at Buy with an $80 target.
What to watch
The Q4 BLA filing for Zenkuda is the make-or-break regulatory step, with the FDA’s verdict on the durability claims to follow. PEAK MESI data in Q4 is the next pipeline catalyst, and traders should expect continued volatility plus active options hedging around these binary events.

Does non-inferiority plus durability win share from Eylea, or is the bar for displacement higher?
